Cognitive / Nootropic
Dihexa
PNB-0408 · N-hexanoic-Tyr-Ile-(6) aminohexanoic amide
What it is
Dihexa is a small molecule developed at Washington State University as an angiotensin IV analog. Reportedly ~7–10 million times more potent than BDNF at promoting synaptogenesis (formation of new synapses between neurons). One of the strongest known cognitive enhancers in animal models.
How it works
Activates the hepatocyte growth factor (HGF) receptor c-Met, triggering intracellular signaling that builds new synaptic connections. Crosses the blood-brain barrier readily. Effects are durable — new synapses persist after dosing stops.
Benefits
- Massive synaptogenesis — new neural connections
- Memory and learning enhancement
- Possible reversal of cognitive decline (Alzheimer's research)
- Long-lasting effects (synapses persist)
- Crosses blood-brain barrier orally
Timeline
- Day 1–3
- Subtle clarity changes.
- Week 1–2
- Memory and learning improvements.
- Week 4–6
- Cumulative cognitive benefits.
- After
- Effects persist due to durable synaptic changes.
Dosing & titration
Standard dose8–25 mg orally or sublingually daily
Cycle length2–4 weeks; long cycles uncommon
TimingAM with food
When to titrate upLimited human dose-response data. Start at low end.
Side effects & risks
- Limited human safety data
- Possible cancer-risk concerns due to growth-factor-like activity
- Some users report headaches, anxiety, restlessness
- Theoretical risk: c-Met activation is cancer-relevant
Cancer caveat: c-Met activation is implicated in tumor growth. Long-term safety data is essentially nonexistent in humans. Use with caution and short cycles only.
Typical price
$80–$160/moCapsules from research suppliers. Quality varies wildly.
